چکیده

Hydrogen peroxide reacts with ferrous horseradish peroxidase and converts it to oxyperoxidase in a sequence of two reactions. The first is the reaction of ferrous peroxidase with HzOz to form Compound II; the second is the reaction of Compound II with HzOz to form oxyperoxidase. Both reactions follow second order kinetics, being first order with respect to each of the reactants. They proceed without detectable intermediates, and, therefore, appear to be single step Z-electron oxidations.

Hydroxyl-radical production in physiological reactions. A novel function of peroxidase.

Peroxidases catalyze the dehydrogenation by hydrogen peroxide (H2O2) of various phenolic and endiolic substrates in a peroxidatic reaction cycle. In addition, these enzymes exhibit an oxidase activity mediating the reduction of O2 to superoxide (O2.-) and H2O2 by substrates such as NADH or dihydroxyfumarate. Here we show that horseradish peroxidase can also catalyze a third type of reaction tha...

Hydrogen peroxide-independent generation of superoxide by plant peroxidase: hypotheses and supportive data employing ferrous ion as a model stimulus

When plants are threaten by microbial attacks or treated with elicitors, alkalization of extracellular space is often induced and thus pH-dependent extracellular peroxidase-mediated oxidative burst reportedly takes place, especially at the site of microbial challenge. However, direct stimulus involved in activation of peroxidase-catalyzed oxidative burst has not been identified to date. Here, w...

Structural insights into the effects of charge-reversal substitutions at the surface of horseradish peroxidase

Horseradish peroxidase (HRP), has gained significant interests in biotechnology, especially in biosensor field and diagnostic test kits. Hence, its solvent-exposed lysine residues 174, 232, and 241 have been frequently modified with the aim of improving its stability and catalytic efficiency. In this computational study, we investigated the effects of Lys-to-Glu substitutions on HRP structure t...

Stabilization of the substrate reaction of horseradish peroxidase with o-phenylenediamine in the enzyme immunoassay.

When the horseradish peroxidase reaction is stopped with acid, the decay of unconverted hydrogen peroxide is responsible for the further oxidation of o-phenylenediamine. This leads to a time-dependent flattening of the standard curve in the enzyme immunoassay, after the reaction has been stopped. Addition of reducing agents, such as sulphite ions, to the stopping solution, prevents the further ...

Characterization of the N-Demethylation Reactions Catalyzed by Horseradish Peroxidase*

The hydroperoxide-supported N-demethylation reactions catalyzed by horseradish peroxidase have been characterized in detail. The ethyl hydroperoxide-supported N-demethylation of N,N-dimethylaniline by horseradish peroxidase resulted in the formation of equimolar amounts of N-methylaniline and formaldehyde with no other products detectable by high performance liquid chromatography analysis of th...
